Haunted Mansion is the first thing we ride; it's a family tradition now. Gotta love the Tower of terror, though.

Haunted Mansion for me (and we cannot ride it any less than 3 times )

Beyond that our other can't miss attractions are:

    Spaceship Earth
    Test Track
    O' Canada
    Small World
    Pirates
    TTA

And while everyone else is enjoying(?) Pooh's honeypots, I close my eyes and imagine that I'm still riding in Mr. Toad's Motorcar!

It's hard to name just one. At MK - almost everything is a don't miss - either because of the awesome-factor or the nostalgia factor, so I can't even think about this question for MK rides.

Otherwise, the one I would be the most disappointed to miss would be Soarin'

    Oh, this is hard!

MK--Pirates, IASW, Peter Pan, BTMRR, CoP, Dole Whip Float!

EC--SE for my sister, Maelstrom for me, Soarin' and Test Track for both

DHS--ToT for me, Little Mermaid for everybody

AK--EE, Safari (my sister is obsessed with zebras!)

    and of course the monorail!

    Ours has been:
DD: Little Mermaid Show at DHS
    DH: Star Tours
me: Dole Whip (okay, not necessarily an attraction, but absolutely will not miss out!)

    Thanks!
Well you make it hard by saying "ONE" ... especially at MK for me, but IF I have to choose, it would be Haunted Mansion for sure.

Some of my favorite attractions, I am ok with passing depending on my schedule (or the weather, in Splash Mountains situation!) but I would never miss Space Mountain.
