DME with one nights stay

My In-laws may spend one night at Disney with us (it's my FIL b.day and he'll get in free). If they stay on-site can they use DME to get back to the airport?

They will have their own room.

Yep...they should make the Magical Express reservation at the same time that they make their hotel reservations...if they've already made the hotel they can either call Disney directly and they will connect them with the ME phone # (I can't find it right now, and I normally use Magical Journey's and they handle the ME reservation).
